Film ini menceritakan sebuah kisah dari kitab Perjanjian Lama. Di Persia, seorang gadis Yahudi nan jelita bernama Hadassah tinggal bersama Mordechai, famili yang mengangkatnya sebagai anak. Hadassah berteman akrab dengan seorang pria bernama Jesse dan mereka berdua bercita-cita untuk pergi ke kota Yerusalem. Namun semua itu harus berakhir ketika raja Xerxes Agung mengadakan sayembara untuk mencari permaisuri. Hadassah harus menjalani seleksi di istana, sementara Jesse dan para pemuda lain dijadikan kasim di istana.
Sebelum pergi, Mordechai meminta Hadassah untuk merahasiakan identitasnya sebagai Yahudi dan memberinya nama ‘Esther’ yang lebih berbau Persia. Setelah melalui berbagai kejadian, sang raja jatuh cinta dan memilih Esther sebagai sang permaisuri.
Di istana, Esther berbahagia dengan suaminya hingga suatu saat ia menyadari sebuah plot jahat yang direncanakan oleh Haman orang Agagit yang bertujuan memusnahkan kaum Yahudi di Persia. Esther pun berusaha mencegahnya, hingga ia rela memberikan nyawanya sebagai taruhan.
